1. EXECUTIVE SUMMARY

  • Action objectives during the reporting period

The main objective of GrapeGen06 is to contribute to the successful long term preservation of the Vitis genetic resources for the use of future generations. By ensuring an optimized characterization of these resources, we will promote their use and ensure the competitiveness and robustness of the European wine sector.
Five main goal were defined within the project (see below for their description). The project being planned for 4 years, each of these general objectives will not be achieved during the first year. Furthermore, as in each project, Year 1 is usually the "starting" year, and one of the main "side objective" is to ensure a good running of the project, that all partner have understood the planned work and are well "on their way" to achieve the global objective of the project.
Objective 1:Monitor still existing Vitis germplasm in Europe, including Eastern countries
For this general objective, the intermediary objectives during Year 1 were :
For cultivated material maintained in collections:
  • To update data already recorded in the database developed during project Genres081 ( for 10 partners
  • To enter passport data for the 23 other partners that joined the European genetic resources community within this new project.
For old autochthonous cultivars in on-farm conservation
  • To identify growers maintaining interesting material on farm for several old autochthonous cultivars and interested in collaborations
  • To confirm the identity of the material
For wild material in-situ :
  • Before to monitor the existing material, survey had to be done. Because some partners were not familiar with this work, we had to write and agree on a common protocol
  • Start some surveys in order to identify the individuals still existing in wild conditions in the different countries of the partners.
Objective 2: Describe this germplasm, using common morphological, agronomic, and molecular descriptors and linking them to a unique web-database ()
Again, this aspect will take 4 years to be achieved. The intermediary objectives in this points were:
  • To agree on common practices for the different analysis and develop some standards enabling the easy comparison of data between labs
  • To agree on the minimal list of material to be analyzed during the project , even if this list may be extended during the course of the project if work is going faster than planned.
  • To start the analysis of some accessions by molecular & morphological methods
Objective 3:Implement true-to-type identification of varieties decreasing the number of errors, homonyms and synonyms affecting the European collections.
In order to publish a precise catalogue of the European genetic resources available in the different germplasm collections, we have to ensure that the possible homonyms or synonyms or questionable accessions in the collections are identified and compared.
As this work need the development of tools and the availability of morphological and SSR data, the work planned in year 1 was :
  • To implement the database with new tools enabling easy comparison of data
  • Draw a first list of questionable cultivars that will need to be analyzed in more details during the following 3 years

  • Action objectives during the reporting period (suite)

Objective 4: Promote an overall optimized conservation scheme for the safe conservation of the Vitis germplasm including Vitis sylvestris currently threatened of extinction, and involving several conservation means (ex-situ collections, cryopreservation and on-farm conservation by farmers and professionals)
This action was dependant on the accumulation of data (identification of wild individuals or of on-farm vineyards, trueness-to-type of the individuals,…) before to draw a global scheme. The safe conservation of germplasm nevertheless need the development of methodologies for cryo-preservation. Development or adaptations of technologies for cryoconservation of seeds, pollen and buds were thus performed.
Objective 5 : Ensure the overall accessibility of the information by the mean of diverse media (scientific papers, technical papers, general public publications, websites, ..) and ensure the availability of healthy material for the use by European growers
For this aspects, work will be done all along the project and when data will be available. However, several specific aspects were necessary in order to help achieve these objectives and were performed during this first year :
  • Development of a website for the project including a private access website for easier management of documents.
  • Improvement of the existing web database for easier use
Globally, these 5 objectives were declined into 7 work packages. Objective 1 and 2, correspond to Task 1 of the project (Acquisition of plant materials and data) including WP 1 to 4. Objective 3, correspond to WP 5, objective 4 to WP 7 and objective 5 correspond mainly to WP6, but also some aspects of WP2 and 3 for the sanitary aspects.
  • Key results achieved and outputs

Results and outputs of the projects will be given according to the general objectives presented previously. Nevertheless, since the detailed results will be given by WP (see part 2), this presentation will somewhat be different. Individual contribution of the partner were not added to this documents but will be available on request from the coordinators.
In relation with Objective 1
Result 1: Adaptation of Multi Crop Passport Descriptor (MCPD) to Vitis specific purposes
Result 2: Identification of 31 growers willing to participate to on-farm analysis and of 113 cultivars to be analyzed
Result 3:Confirmation of identity of the cultivars in the on-farm analysis (5 partners out of 15)
Result 4: Definition of the inventory protocol (D16).
Result 5: Identification of more than 100 sites with V. sylvestris material (D17).

During summer 2007 all the partners involved in this sub-task received the DNA extracted by INRA (P0) from 47 reference cultivars according to the distribution list agreed during the first meeting (11 or 12 samples per partner, Annex 2).
Different apparatus were available in the partners’ labs for the genotyping of cultivars at SSR loci (polyacrilamide vertical electrophoresis systems and silver staining, ALFexpress II, ABI 3130) and we decided to compare them.
Partners P0, P1,, P3, P6, P7, P8 & P13 analyzed all the cultivars at the 9 SSR loci and delivered the data to the coordinators. Partners P5, P9 and P20 used only 6 core (Genres) loci to analyze 4, 35 and 12 reference cultivars respectively. The analysis of Partners P2 , P4 and P21 are in progress. (Annex 1).
Generally, there is sufficient amount of data for validation and definition of reference alleles and cultivars. As for previous analysis during the previous project (Genres081) several discrepancies in genotyping some specific SSR loci were detected. The identification of the reference alleles is not finished yet, because of some misunderstanding of the codes.
  • Sub Task 1.2: Microsatellite analysis of chosen accessions

All the Partners involved in WP1 agreed to provide molecular data at 9 SSR loci for a defined number of cultivars. The number of cultivars to be analysed by each partner was reported in Annex 1. Some partners already provided the list of the cultivars to be analyzed, while some are still in the process of drawing it.
Six out of 21 partners delivered 25-50% of the anticipated raw SSR data for all 9 agreed SSR loci, while 10 others performed genotyping on 6 core SSR loci up to now. Five partners are in the process purchasing labs equipment, optimizing reactions or repeating the preliminary results.

Generally, 10-50 % of genotyping work is completed but data adjustment and quality control remains to be done and cross-validated in the next years.
  • Sub Task 1.3: Encoding of data (already available and new).

This sub-task is very dependent on the previous two ones.
P0 provided the allele codes and the excel macro which allows the coding of individual partner’s data. Generally, most of the partners were able to run the macro provided by the partner P0 and to turn raw data into coded allele values. However, the discrepancies in the allele detection among different labs using the “same DNAs” and the difficulties in using the Macro delayed the work of few partners.
  • Sub Task 1.4.: Coordinated microsatellite analysis as needed within WP5

The project anticipates the necessity of genotyping some cultivars originating from various sources and across several partner labs in order to define the true-to-type status of some accessions. Partners are allowed to raise the “question of identity or homogeneity” for some cultivars and to invite other partners to solve it (see WP5). At this moment, there was no any activity in this sub-task but this is probably going to become an issue in next years after national data will be merged into master database.

Data acquisition for the SSR-marker database within the EU-Vitis database
  • Conclusions

Within this WP, the acquisition of the row data is progressing well : Even if a little slow for the reference set and even if some partners are still purchasing equipment, more than 20 % of the row data is available. We are thus not anticipating any problems yet.
The encoding and transfer to database will need the development of the common alleles. This aspect is progressing but is a little behind schedule. Particular attention will be given to this aspect at the beginning of 2008, in order to be sure to get the reference at the latest by june 2008. A demonstration of the macro will also be proposed in Hungary, during the meeting for those who did not understood it. Nevertheless, this has no effect on the acquisition of new data, and will have no effect on the transfer of data to the database since this transfer will not be possible before the end of 2008 (see WP6).

A summary list of old endangered and autochthonous cultivars that will be studied has been elaborated (Annex 3 ).
The updated list of I and II descriptor codes (OIV-IPGRI) has been provided. A model format for facilitating data circulation and submission by each partner to the database was also prepared.
Grape descriptor definitions have been considerably improved and harmonised between OIV, IPGRI and UPOV. The harmonisation started in 2002 and finished in 2007. Emphasis was given on easiness in recording, objectivity and minimal modification sensibility. 48 descriptors were selected from the 151 OIV descriptors published in June 2007 in order to use in the GrapeGen06 project.
On behalf of the International Organization of Vine and Wine (OIV) Partner 1 has coordinated the harmonization of the UPOV, Bioversity and OIV descriptors for grapevine varieties and Vitis species. In June 2007 the 2nd edition of the descriptor list has been accepted by the OIV General Assembly. For this reason partner 1 has been involved in the compilation of the primary and secondary descriptors to be applied in GrapeGen06. Partner 1 provided the excel file format for data recording. A standardized format is a prerequisite for acquisition of data in the European Vitis Database.
A common database has been organized with the data generated by every partner. The work done by partners has been summarized in Annex 3. Globally, more than 1/4th of the data were collected (full description of 200 cvs out of 700).
  • Sub Task 2.2: Realisation of the pictures of cultivars

The main organs of every cultivar selected for the ampelographic study will be photographed during the project.
Detailed results of the different partners are presented in annex 4. Globally, this sub-task is only at it's beginning, few partner having started with the pictures. But it is no surprising way since selection of the cultivar was only made during the first year and analysis start with ampelography. Usually pictures are taken later on .
After discussions between partner P0, P1 and P3, a common system for labelling has been proposed, taking into account the link with the European Vitis Database.
For Partner 23, pictures were taken at Vassal domain, on 20 native Georgian cultivars similar to those present in partner's 23 collection.
  • Sub Task 2.3:Characterization of the sanitary status of cultivars

A list of a new set of analysis for phytosanitary test was made: The minimal list of virus to be tested are Grapevine fanleaf virus (GFLV) , Grapevine leafroll-associated virus type 1 (GLRaV-1) and type 3 (GLRaV-3) using the ELISA method (agreed during workshop in Versailles).
Results achieved
Only partners 3, 12, 16 have started this sub-task by Elisa testing (Annex 3). Because of unclear results, partner 3 (IMIDRA, Spain) will need to repeat some of the analysis. In order to maintain the virus free state of 8 cultivars, partner 16 (ARI, Cyprus) maintained them in two fully insect-proof screen-houses at Zygi experimental station, as pre-basic propagation material.
Partner 20. FAZ (Croatia) has planned to collect cuttings for the virus tests during the wintertime. But, ELISA tests conduction will depend on available money.
  • Sub Task 2.4.: Preparation of the catalogue of autochthonous cultivars

The catalogue of autochthonous cultivars will be elaborated during the project with the compiled results: ampelographic descriptions, photos and phytosanitary status. This sub-task will be thus started later on during the project.

Title: Acquisition of primary and secondary descriptor data of old, endangered and autochthonous grape varieties
  • Conclusions

The partners involved in this WP agreed on the list of descriptors to be analyzed. The work started this year and sub-task 2.1 is running well since 1/4th of the data was collected. No problem is anticipated for this task. A training course will also be performed in 2008 (before the meeting) to ensure homogenisation of the data. Subtask 2 and 3 are behind schedule so particular attention should be given to these points next year.
