1Generation of dopaminergic neurons from induced pluripotent stem cells using B-lymphocytes

Objectives: The study aimed at generating patient-specific pluripotent stem cells as it has the potential to accelerate the implementation of stem cells for clinical treatment of neurodegenerative diseases like Parkinson’s.
Methods: B-lymphocytes isolated from spleens of rats were reprogrammed for generation of induced pluripotent stem cells (iPSCs)by inducing expression of the transcription factors KLF4, OCT4, SOX2, and C-MYC. The expression of the transcription factors in the reprogrammed cells was analysed by reverse transcription-polymerase chain reaction (RT-PCR). The resultant iPSCs lines were subjected to the differentiation medium to obtain neural stem cells (NSCs). NSCs differentiated into dopaminergic neurons were examined for immunocytochemical expression of TH marker.
Results: Agarose gel analysis following RT-PCR revealed distinct expression of KLF4, OCT4, SOX2, and C-MYC. By Immunocytochemistry the NSCs generated from iPSCs were found to express neural and glial markers such as Nestin, GFAP and OCT4. Also, the NSCs differentiated into dopaminergic neurons were found to be TH positive.
Conclusion: These results suggest any somatic cell could be reprogrammed to a iPSC and these iPSCs could have a clinical implication in degenerative diseases
