NanoDrop user guide

1-  Turn on the computer.

2-  Launch the NanoDrop 2000 software.

3-  Select the right assay (Nucleic Acid).

4-  Wavelength verification step. Remove the kimtech under the arm of the nandrop and click OK.

5-  Select the right type of sample (DNA, RNA, ssDNA, …) (colored top right field) and the absorption coefficient will be modified accordingly.

6-  Before any measurement, ensure that the box “add to report” (top left) is checked in order to save the totality of your data.

7-  Load 1.5 – 2 mL of ddH2O or DEPC H2O on the pedestal reading surface. Put the NanoDrop arm GENTLY down and click on “Blank” (top left).

8-  Wipe the drop from the pedestal and arm surfaces with a clean kimtech.

9-  Enter your sample ID.

10- Load 1.5 – 2 mL of your sample on the pedestal reading surface. Put the NanoDrop arm GENTLY down and click on “Measure” (top left).

11- Repeat steps 8 to 10 (cleaning, loading and reading) for all your samples.

12- Load 1.5 – 2 mL of ddH2O or DEPC H2O on the pedestal reading surface. Put the NanoDrop arm GENTLY down and read its absorbance as a sample by clicking on “Measure” (top left). You expect to have a 0ng/mL concentration and no absorbance peak. This step is to confirm the validity of your measurements.

13- Wipe the drop from the pedestal and arm surfaces with a clean kimtech and clean the NanoDrop using a kimtech moistened with ddH2O or DEPC H2O.

14- Click on the “report” bar (bottom left).

15- Click on the “Export” button (top left).

16- Save your data in a *.XML format file on a USB storage device.
