Movie legends

Montage movies of tetO-lacO and lacO-tetO tandem arrays. Replication was blocked at tetO for 70 min in liquid culture, followed by an additional 30 min on the slide before imaging. Images were taken every 3 min. Top left panel, overlay of TetR-YFP (green, block) and LacI-CFP (red) channels; top right, TetR-YFP (green, block) channel; bottom left, LacI-CFP (red) channel; bottom right, phase contrast (blue).

Movie S1. Time-lapse analysis of tandem array tetO-lacO. LacO (red) is immediately downstream of the block, tetO array (green).

Movie S2. Time-lapse analysis of tandem array lacO-tetO. LacO (red) is immediately upstream of the block, tetO array (green), which allows the lacO to be replicated, thus giving a much brighter CFP signal than in Movie S1.
