Buffer optimization for NMR samples using microprobe

  1. Buffer preparation: MJ001-MJ012
  2. Purify protein with Ni-NTA affinity column, optionally followed by gel filtration chromatography.
  3. Concentrate protein samples to ~ 1.0 - 1.5 mM in elution buffer
  4. Change to buffer MJ001-MJ012 using desalting plate
  5. Measure concentration using nanodrop
  6. Transfer samples to microtube (1 or 1.7 mm)
  7. Monitor the samples and recorder the status
  8. Store the samples at 4 C for one week
  9. Monitor the samples and record the status again
  10. Direct the samples to NMR screening
