Agilent Technologies 1200 Series Analytical HPLC

The Agilent Technologies 1200 series HPLC was first released in 2007. It is an upgrade of the 1100 series HPLC consisting of upgrades to XXXX……………….

Comparison between the Agilent 1200 and Varian 920-LC

Compared to the Varian 920-LC the Agilent 1200:

·  Standard autosampler cannot accept wellplates and the microplate 1200 series autosampler cannot accept 1.6 mL HPLC vials. The Varian 920-LC autosampler is capable of injecting from 1.6 mL vials, 96 well microplates or 384 well microplates.

·  Is limited to 100 HPLC vials in the autosampler compared to 200 x HPLC vials in the 920-LC

·  The standard 1200 series autosampler has a large 300 mL delay volume compared to the 200 mL on the Varian 920-LC automated sample handler.

·  Variable wavelength UV Vis detector is a single l detector only. The Varian 920-LC has dual l capabilities with real-time purity checking.

Agilent claims the 1200 series quaternary pump

Offers flexibility for solvent selection and automated method development and is ideally suited to the multi-method high throughput laboratories.

·  Provides convenient front access to all maintenance parts for quick replacement without having to disassemble the instrument

·  Early Maintenance Feedback (EMF) continually tracks long term usage with pre-defined limits and feedback messages if the limit is exceeded

·  Full programming capability such as intelligent method sequencing which includes built-in automated shutdown methods to reduce solvent costs.

Note for the Agilent 1200 series quaternary pump system to work properly a 1200 series 4 channel vacuum degasser is required. To safely mount the solvents, a

What Agilent says about the 1200 series Autosampler

The standard 1200 series autosampler holds only 100 industry-standard vials.

Claimed features of the 1200 Series Autosampler are:

·  Reproducible injections from 0.1 to 100 mL

·  Carryover typically < 0.1% without needle wash and < 0.05% using needle wash

·  Overlapped injections to improve sample throughput

The 1200 series high performance autosampler has the following extra capabilities

·  2 x 96 (or 384) wellplate capacity

·  2 x 54 position HPLC vial racks

·  Reduced delay volume for working with fast gradients by bypassing the injection valve after an injection is made.

What Agilent says about the SSI Chromatography Data Software

·  It’s quick and easy to store and retrieve chromatography data using an embedded, relational database.

·  Take advantage of more precise peak integration calculations with ApexTrack™ algorithm.

·  Easily control Waters ACQUITY UPLC™ and Alliance® HPLC systems as well as third-party HPLC and GC systems.

·  Enhanced detection capabilities are available when using Waters mass detectors or the Waters 2996 Photodiode Array Detector.

·  Scalable, modular architecture allows seamless growth from a single workstation to an enterprise-wide system.

·  Designed for a data-secured regulatory lab environment – three clicks to compliance.

What Agilent say about the 1200 Series VWD

Agilent claim a simplified optics design is optimized for high sensitivity and low baseline drift

·  Programmable wavelength selection for sensitivity and selectivity

·  Minimum baseline noise and maximum stability for lowest detection limits

·  A wide linear range of 2 AU for simultaneous quantitation of major and minor compounds

·  A choice of flow cells including

o  10 mm path length 12 mL dead volume and 40 bar pressure rating

o  4 mm path length 1 mL dead volume and 40 bar pressure rating

o  High pressure 12 mL volume 10 mm, 400 bar pressure rated path length flow cell

·  Holmium oxide filter for automatic wavelength verification

The 1200 Series VWD has a D2 lamp only and wavelength range limited to a maximum of 600 nm, severely limiting its capabilities in the visible wavelength range. It’s 13 Hz data acquisition rate limits it use for high resolution high throughput type applications. A noise specification of + 7.5 x 10-6 Au for the 1200 Series VWD is poorer than the + 5 x 10-6 Au of the UV Vis detector in the 920-LC which ensures superior sensitivity performance of the 920-LC.
