Standard Operating Procedures for PHORBOL ESTERS and PHORBOLS
#1 /Process
Handling and weighing pure chemical to prepare administration dosages for induction of tumors.#2 /
Hazardous Chemicals/Class of Hazardous Chemicals/Signs & Symptoms of Exposure
Phorbol and phorbol esters including phorbol myristate acetate. Very Toxic to Harmful. May be fatal if swallowed. Readily absorbed through skin. Irritating to skin, eyes, and mucous membranes. Phorbol and phorbol esters as a class are some of the most potent tumor promoters known to man. Some phorbol esters enhance tumor promotion of known carcinogens ten fold. Experimental evidence indicates PMA to be carcinogenic, teratogenic, and mutagenic.#3 /
PPE - Personal Protective Equipment
Wear two layers of chemical resistant glove like disposable nitrile, safety goggles, and laboratory coat. Material is readily absorbed through skin.#4 /
Engineering/Ventilation Controls
Use with safety shower and eyewash nearby. Use only in a chemical fume hood until diluted into liquid.#5 /
Special Handling Procedures and Storage Requirements
Read Material Safety Data Sheet prior to use.Use extreme caution when handling these compounds with carcinogenic compounds.
Do not breathe dusts. Do not get in eyes, on skin, on clothing.
Keep in tightly closed container. Store in a cool dry place at -20°C.
Protect from light. Avoid strong bases and strong oxidizing agents.
Order as small as quantity needed.
Wash thoroughly after handling.
#6 /
Spill and Accident Procedures
If outside the hood, evacuate unnecessary personnel, contact EH&S as self-contained breathing apparatus is required. If in hood, then gently sweep up, place in a bag and hold for waste disposal. Avoid raising dust. Ventilate area and wash spill site after material pick up is complete.#7 /
Waste Disposal
Dispose of all items contaminated with phorbols as hazardous waste through EH&S, including empty source vials & containers.#8 /
Approval Required
Supervisor approval required for first use.#9 /
Decontamination Procedures
After removing bulk material via absorption or gentle sweeping, wash surfaces with soap and water.#10 /
