Supplementary Fig 1 HPL Optimization and isolation efficiency: (a) Bone marrow-derived mesenchymal stromal cells (BM-MSCs) grown on various concentration of human platelet lysate (HPL) - 5%, 10% 15% and 20% shows the optimum cell yield at 10% of HPL. (b) Plating efficiency of BM-MSC at P0 in four different media. Yield at P0 is significantly higher in LG-HPL (p<0.05).

Supplementary Fig 2 CFU was examined in two different basal media (DMEM-KO and DMEM-LG) having HPL or FBS as supplements. Cultures were stopped and assessed for CFU determination on day 14 by staining with crystal violet. Representative documentation of one replicate out of three of four donors is shown.

Supplementary Fig 3 Immunophenotype of BM-MSCs: The marker profile commonly referred to as characteristic for BM-MSCs was measured by flow cytometry. Green lined histograms shown target antibody reactivity of LG-HPL, red lined histograms indicates LG-FBS and gray histograms represents the corresponding isotype control reactivity (a-h). Similarly orange lined histograms shown target antibody reactivity of KO-HPL, blue lined histograms indicates KO-FBS and gray histograms represents the corresponding isotype control reactivity (i-p).
