Supplementary Fig. 3 RT-PCR detection of mouse Faim and Spag6 gene expression. As a control, parallel reaction was carried out using genomic DNA as starting material. Faim and Spag6 genes are labelled with white or back circles in the same way as in Figs. 1 and 2. We noted that the PCR results are not quantitative measurement of expression level, and merelyprovide the proof that mouse Faim and Spag6 genes are indeed expressed in vivo and thus indicate functions.

Total RNA was extracted from mouse tissues using the NucleoSpin RNA II kit (Macherey-Nagel, Düren, Germany). The DNaseI treated total RNA (2 μg) was primed with oligo-dT primer and reverse transcribed at 42OC for 2 hours using the High Capacity cDNA Reverse Transcription kit (Applied Biosystems, Foster City, USA). 2 μl of cDNA was used in PCR reactions with gene specific primers. To monitor possible genomic contaminations, control reactions were carried out with genomic DNA as a template. The quality of cDNA was verified by amplifying of Atp7a transcripts. The used primers are: 5'-CGTAGCTGTTTGGGACGTAGC-3' and 5'-CTCGTATGCGAAGCCACTTATG-3' for Faim; 5'-TCAGACCCAAGACGTTGCTTTGC-3' and 5'-ACCACAACCTTTGCCAGAAA-3' for Faim-Gm6432; 5'-TTCAGTAAGGTGCTGCCACA-3' and 5'-ATCCTTACTCTCCTGTGAATCTGAGT-3' for Spag6-BC061194; 5'-TTCAGTAAGGTGCTGCCACA-3' and 5'-AGTCCAGCCACTAACCTTAAACTG-3' for Spag6; 5'-CAATAAGATCCTGGCCATTGTGGGG-3' and 5'-CGGTTACCAATGAGGACTTTG-3' for Atp7a.
