SAMPLE SOP ADDRESSING LAB AND ANIMAL FACILITY BIOSAFETY PROCEDURES

Note: if you do not perform any animal work that will affect DLAR put N/A for the “DLAR Staff” SOP

Hazards: The following materials and/or equipment associated with the use of the agent that may present exposure hazards, health hazards, and/or physical hazards.

Herpes Simplex Virus (HSV)

Attenuated and wild-type herpes simplex virus will be used in these experiments. Infectious virus will only be administered intracranially. Thus, after virus administration, animals will be monitored for signs of paralysis, which will denote development of encephalitis. Any animals developing encephalitis will be immediately sacrificed following IACUC protocol. Viral shedding from the animals which do not develop encephalitis will be minimal. Animals which do develop encephalitis are capable of shedding virus, but as the route of infection is intracranial, shedding upon this route of infection is minimal. Furthermore, shedding time will be minimalized, as animals will be sacrificed upon first signs of encephalitis development.

Containment Procedures: The following safety equipment must be used when carrying out this procedure: (chemical fume hood, biological safety cabinet, sealed centrifuge rotors/ covered centrifuge tube buckets, filter cage top, etc.)

DLAR staff: All work with virus-infected animals will be performed in the animal room following standard animal room SOP. All infected-animal work will be performed in biosafety cabinet.

Lab staff: Handling or manipulating agent in lab will be performed in biosafety cabinet. Centrifuged specimens will be loaded and removed inside the biosafety cabinet.

Protective Equipment: The following personal protective must be worn when performing this procedure: (list type of glove, eye protection, lab coat, etc.)

DLAR staff: Personnel will wear gowns, gloves, face mask, hair nets, and footies at all time while in the animal room.

Lab staff: Personnel will wear gowns or lab coats, gloves, at all time while in the lab.

Decontamination/Clean-Up Procedures:

DLAR staff: Biosafety cabinet will be wiped down with Spor-Klenz 1:5 solution immediately following any activity with mice. Hood will not be used for at least 20 minutes post-usage. All waste (bedding, paper towels, etc.) associated with animals will be disposed of in biohazard bags and autoclaved, and disposed of by DLAR procedures for the length of the stay of the animals. Cages will be autoclaved as per DLAR protocol.

Lab staff: Biosafety cabinet will be wiped down with a Spor-Klenz 1:5 solution, or another appropriate disinfectant, immediately following any activity. Hood will not be used for at least 20 minutes post-usage. Any small biological agent spills outside the biosafety cabinet will be covered with a paper towel and then treated with bleach (wear gloves). Let sit for at least 20 minutes and then dispose of into autoclave bag. Spills inside the biosafety cabinet follow same procedure, except use a non-bleach disinfectant.

Waste and Animal Disposal Procedures (bagging, autoclaving, etc.):

DLAR staff: Animal carcasses will be disposed of in marked biohazard bags and placed in carcass freezer per standard DLAR protocols.

Lab staff: Infectious waste, such as tubes, eppendorf tips, and other non-sharp items will be disposed of in infectious waste containers (biohazard/ autoclave bags) within the lab. Used sharps will be placed in a sharps container. Liquid biohazardous waste will be autoclaved, or in the alternative, treated with bleach and dispose of down the sink

Injury/Exposure Response Procedures:

DLAR staff: Any exposed area coming into contact with animals should be immediately washed with soap and hot water (notify supervisor). Area should be observed for 7 days. Any further treatment go to University Health Clinic (4K). Emergencies, please contact WSU Public Safety at 577-2222

Lab staff: Any exposed area coming into contact with biological agent or infected animal tissue, should be immediately washed with soap and hot water (notify PI). Area should be observed for 7 days. Additional Special Handling Procedures, including transport of agents (secondary containment):

DLAR staff: All biosafety procedures are to be followed the entire time the mice are in the animal room.

Lab staff: All biosafety procedures are to be followed the entire time while in the lab. Any transport of biological agents between labs or buildings will be done using sealed secondary containment to prevent accidental spills.
