Phosphate analysis

-Make standard solutions with three replicates at 8 concentrations including blanks

-Make reagent, make 100ml due to 78 samples

-Add 1ml of reagent to each sample and mix

-Leave for 1 hour

-Put in U1800 spectrometer

-Take 2 readings and average

-Record data on sheet

Zooplankton analysis

-Conventional methodology used to count zooplankton i.e. microscope and Bogarov chamber to analyse 500ml samples

Chlorophyll analysis

The chlorophyll bottles were transferred asap to s freezer after collection aboard the Bill Conway on the 8th June; thus preserved the samples. The following morning the samples were ready to analyse as the action of freezing allowed the chlorophyll to be released from the filter into the acetone solution instead of having to centrifuge the sample. Conventional methodology was then used to calculate the concentration of the chlorophyll in each of the samples.

To calculate the chlorophyll concentration in the water bottles:

(6/50) x value given by the flurometer where 50 was the volume of the filtered solution in ml and 6 was the volume of acetone in ml.

NB: need to calibrate the fluorometer would normally use a known concentration of pure chlorphyll a, approximately 90% acetone.

Plan:

-Using Niskin bottles on a rosette water samples were collected so DO, Silicate, Phosphate, and Phytoplankton (Lugols) could be measured.

-Using a closing zooplankton net zooplankton samples were collected

-Using an ADCP water flow would be measured.

-Using a CTD with fluorometer and transmissometer, temperature, depth, salinity, turbidity, and fluorescence by phytoplankton would be measured.

-Using a Secchi disk turbidity would be measured.

On the basis that a single profile using all of the above techniques to sample the water column, would take ~45mins to complete, that Callista would travel ~10knots, and the first station at Black Rock would take ~1hr 30mins to complete including travel from the Dock, timings were estimated.

NB Time = Distance/Speed, where speed is in knots/nautical miles (nm) per hour, and distance is in nm (1 nm = 1 degree of latitude).
