Appendix 1. Human Health and Ecological Reference Values Used in this Study

Human Health Reference Values

The MIAT decision tree requires data on Reference Values (RVs)for human health and ecological effects. The human health effects should be based on the noncarcinogenic endpoints of the compounds. The samples were grab samples; therefore, the most appropriate standards for use in this assessment are acute oral toxicity standards. However, acute oral standards were not available for most of the compounds. In this study chronic standardswere used as a conservative measure of acute RVs. Preference was givento standards or advisory criteria set by the European Union (EU) over those set by individual European countries. Preference was given to enforceable standards rather than guidance criteria and final values over proposed values. When drinking water standardswere considered for use as the basis for the RVs, we excluded standards that are based on economic considerations, analytic considerations, or limitations inwatertreatment technology. Finally, because our analysis focuses on the effects from exposure to the mixture of chemicals from a specific source and not the cumulative risks from all sources of the contaminants, it is inappropriate to include the source-apportionment factor.RVs of four pharmaceuticals (diclofenac, erythromycin, erythromycin+erythromycinH2O, and mefenamic acid) were based on minimum therapeutic dose (MTD), a body weight of 60 kg, and an adjustment factor of 1000 according to Australian guidelines for water recycling[1].A list of the standards and their sources are provided in Tables A-1and A-2.

When human health criteria were not available, the Cramer class portion of the TTC was used to conservatively estimate a safe dose. This was done by assuming anuncertainty factor of 100 and the point of departure is set for each of the three Cramer classes (Price et al, 2007). This approach was applied to 107 chemicals. The approach could not be applied to seven common ions and to four polynuclear aromatic compounds.

Ecological Reference Values

The ecological criteria were based on intermittent standards when they were available. Standards were only acceptedif they are based on acute toxicity values and that were calculated according to the methods outlined in the technical guidance document for deriving environmental quality standards (European Commission 2011) for the MAC-EQS and according to the methods outlined in the technical guidance document of risk assessment ( European Commission 2003) for the PNECintermittent. In cases where such acute data were unavailable chronic standards like the AA-EQS (European Commission 2011) or the PNEC (European Commission 2003)were used. Since the AA-EQS is equal to or lower than the MAC-EQS by definition, this substitution is in line with the precautionary principle.The reason of preferring reference values for intermittent exposure over reference values for chronic exposure is that the exposure datasets, with only one-time measurements do not allow for the calculation of chronic exposures, which would require at least 12 measurements per year. Preference was givento standards or advisory criteria set by the EU over those set by individual European countries. Preference was also given to enforceable standards rather than guidance criteria, and final values rather than proposed values. For some substances (mostly pharmaceuticals), RVs published were not in line with the two above mentioned guidance documents, because higher assessment factors were used, indicating that not all required data were available for the derivation. These values were not considered for the assessment.

In certain cases where the degradation products are missing an RV, the parent compounds RVs were used. A list of criteria and their sources are provided in Tables A-3 and A-4.
