Human hepatoma Cell line, Hep3B was cultured in minimal essential medium (MEM) supplemented with 10 % fetal bovine serum (FBS) and 1% penicillin-streptomycin at 37 oC in a 5% CO2 incubator.

Immunohistochemistry for α-fetoprotein (AFP)

AFP immunostaining on the liver section was performed using the monoclonal mouse antibody against AFP (Novusbio, Littleton, CO, USA), with their respective antibodies. The antigen-antibody complex was visualized with avidin-biotin-peroxidase complex (ABC) immunostaining (Vector laboratories, Burlingame, CA, USA) by 3,3-diaminobenzidine (Zymed laboratories Inc., San Francisco, CA, USA). The sections were then counterstained with Mayer’s hematoxylin or methyl green.
