Deletion of TGA stop codon in exon 17 of the major DDX3X transcript class in its 3´UTR splice variants III-VI modify and extend their putative open reading frames (ORFs) for protein coding in the C-terminal regions when starting at the ATG translation initiation codon in exon 1 (Fig.3).

If translation would occur, variant III would encode a protein isoform of the major DDX3X protein where the last 26 amino acids (aa) are replaced by a novel 97 aa long sequence, in variant IV and V by a novel 10 aa long sequence, and in variant VI by a novel 4 aa long sequence (novel peptides are coloured light grey in the DDX3X protein sequences given below). Molecular weights expected for these DDX3X protein isoforms would be 81.49 kDa (III), 71.49 kDa (IV and V) and 70.85 kDa (VI), respectively.
