TITLE: EFFECT OF AQUEOUS LEAF EXTRACT OF BRYOPHYLLUM PINNATUM ON THE BRAIN OF STROKE INDUCED MALE ALBINO WISTAR RAT.

ABSTRACT

This study was carried out to evaluate the effect of aqueous leaf extract of BryophyllumPinnatum on the histoarchitecture of the cerebral cortex of stroke induced male albino Wistar rats. Fourty (40) healthy male albino wistar rats of weight ranging from 200grams to 360grams, were used for this study, bred and housed in the preclinical animal house, Faculty of Basic Medical Sciences, University of Port Harcourt. The animals were kept and nurtured under laboratory conditions, temperature, humidity, and light and were allowed free access to food and water ad libitum.The animals were of four groups and ten (10) animals per group. Group1 was the control group. They were given distilled water. Group 2 was the Ischemic control group. Ischaemia was induced in this group, and only distill water was given to this group without tratment. Group 3, was an Ischemic group, that was treated with 200mg/kg of BryophyllumPinnatum.Group 4, was an Ischemic group, that was treated with 400mg/kg of BryophyllumPinnatum.The administration was done intraperitoneally for two(2) weeks after which, the animals were made to undergo stroke induction by occluding their common carotid artery.The mean and standard deviation of theneurological deficits were evaluated for the various groups andwas analyzed using a one way ANOVA followed by a Post hoc (Duncan test).It was observed that there was a Significant difference between the control and ischemic control in neurological deficits (p<0.05).The Photomicrograph of brain sections of theNormal control group depicted a normal cytoarchitecture ofbrain tissue and neuronal cells with normal nucleus. For the Ischamic control group, it showed a marked congestion of blood vessels, excessive degeneration of neuronal cells , excessive vaculation and necrosis of neural cells. Pretreatment with 200 mg/kg bryophyllumpinnatum showed mild congestion , mild degeneration of neuronal cells, and mild vaculations. Pretreatment with 400 mg/kg bryophyllumpinnatum showed neuronalcells with normal nucleus. It was also observed that the effect of bryophyllumpinnatum on the brain was dose dependent. There was a reversal of the congestion of blood vessels, congestion and necroticdegeneration of neuronal cells compared with Ischemic control group. knowledge gained from this work will be useful in the prevention and management of stroke.

Okoseimiema, Sonny Clement is a lecturer in Anatomy Department, University of Port Harcourt in Nigeria. He is a Ph.D student in University of Port Harcourt, carrying out his research in Neuroscience. His MSc work was on Radiological anatomy of the pelvic region of the Ijaw Ethnic Group in Nigerians. He has Nine publications internationally. He is currently the National Assistant secretary of (SECAN) Society of Experimental and Clinical Anatomist of Nigeria. He has attended about ten conferences for the past four years, and has presented about eleven papers. He has passion for research and desire to grow in research and contribute his quota to the body of knowledge and for the good of humanity.
