Table S1 The nutrient components of the basal diet of mice.

Nutrient component / Proportion
Water / 8-10 %
Crude protein / 20-25 %
Crude fat / 4-5 %
Crude fiber / 4-5 %
Crude ash / 6-8 %
Lysine / 1.20-1.42 %
Methionine + cystine / 0.78-1.10 %
Tryptophan / 0.27-0.34 %
Calcium / 1.2-1.8 %
Phosphorus / 0.8-1.2 %
Vitamin A / 12500-15000 IU/kg
Vitamin D / 1250-1500 IU/kg

The basal diet contained flour, wheat bran, yeast extract, cornmeal, bone meal, sorghum powder, fish meal, salt, soybean powder, eggs, cod liver oil and multivitamins for mice. And the proportion of every nutrient component was given as above.

Table S2 The effect of MAP on body weight, colon length, colonic tumor formation in mice treated with DMH/DSS in the 20th week.

Group / N / Body Weight (g) / Colon Tumor (%) / Colon Length
(cm)
Control / 10 / 45.0 ± 1.6 / 0/20 (0%) / 14.5 ± 0.9#
DMH/DSS / 20 / 43.3 ± 2.9 / 18/20 (90%) / 13.1 ± 1.2*
DMH/DSS+MAP (2.5%) / 20 / 45.1 ± 3.6 / 5/20 (25%) / 14.3 ± 1.0#
DMH/DSS+MAP (5%) / 20 / 44.8 ± 2.5 / 3/20 (15%) / 14.9 ± 0.7#
DMH/DSS+MAP (10%) / 20 / 45.1 ± 1.6 / 1/20 (5%) / 15.2 ± 1.3#

$Mean ± SD. *P<0.01 vs control; #P<0.01 vs DMH/DSS; A total of 100 mice were sacrificed in the 20th week after saline or DMH/DSS treatment. MAP treatment was initiated in the 9th week after the mice treated with DMH/DSS.

Fig. S1. Result of high performance liquid chromatography. The equation of standard curve was got using the Longzhida workstation (Beijing, China): Ln(Mn, weight-average molecular weight)=B0+B1×t+B2×t2+B3×t3 (B0=13.35545, B1= -0.6415637, B2,3=0, “t” stand for time, R2=0.9952). Accordingly, the molecular weight of MAP (modified apple polysaccharides) was about 1000~3000 Da.

Fig. S2. Results of high performance liquid chromatography. (a) The chromatogram of the mixture of monosaccharide-derivatives: 11-phenyl-3-methyl-5-pyrazolone (PMP); 2 mannose; 3 buckthorn; 4 glycuronic acid; 5 galacturonic acid; 6 glucose; 7 galactose; 8 xylose. (b) The chromatogram of MAP (modified apple polysaccharides).Using the chromatogram of the mixture of monosaccharide-derivatives as a standard, MAP was mainly composed of glycuronic acid and galactose and glucose.
