6. BRIEF RESUME OF THE INTENDED WORK:

6.1 NEED FOR THE STUDY:

Rasa Shastra deals with knowledge of Alchemical and Pharmaceutical processes such as Samskaras of Parada, Shodhana,Jarana,Marana etc processes of different Metals,Minerals, etc.substances.

In this current era Indian Pharmaceutics has given Ayurveda a complete novel healthy look.The innate qualities like quick action ,lesser dose,tastelessness,prolonged shelf life and better palatability of Rasaushadhies have helped them to conquer the demand of patients as well as pharmaceutical proprietors.

Panchanana Rasa is an unique herbo-mineral formulation explained in Rasendra Sara Sangraha in Jwara Chikitsa Adhyaya. It a is potent formulation having Hingula, Gandhaka, Tamra, Vatsanabha and Maricha given bhavana with Arka jala which is proven efficient on Jwara.

Pyrexia can be defined as regulated elevation in the body temperature above the customary set point of the hypothalamic thermostat of all the manifestation of infection in the human host1.

Jwara is the disease which is given prime importance in most of the Classics. According to Acharya Charaka, Santhapa is the cardinal feature of Jwara.2

There is no work carried out on Panchanana Rasa so far and its experimental evaluation on Anti Pyretic activity also.

Hence present study is planned to evaluate ‘Pharmaceutico-analytical study of Panchanana Rasa experimentaly in Albino Rats for its Anti Pyretic effect.

6.2 REVIEW OF LITERATURE:

Review of Literature includes thorough screening of Classical Ayurvedic Literature such as Rasendra Sara Sangraha, Charaka Samhita,Rasa Ratna samuchaya,Yoga Ratnakara, Rasa Tarangini,Modern text,Journals etc,regarding the disease and the drug.

1.  Drug Review:

The formulation Panchanana Rasa is taken from the Classical text Rasendra Sara Sangraha, Jwara Chikithsa Adhayaya

Hingula [1 part] ,Gandhaka [3 part],

Tamra [2 part],Vatsanabha [2 part],

Maricha [4 part] and Arka Jala[bhavana] [sufficient quantity].

6.3 AIMS AND OBJECTIVES OF THE STUDY:

1.  To prepare Panchanana Rasa according to Classical method.

2.  To do Physico-Chemical analysis of Panchanana Rasa.

3.  To evaluate the Anti-Pyretic effect of Panchanana Rasa experimentally.

6.4 HYPOTHESIS:

H0 : Panchanana Rasa will not be having Anti-Pyretic activity on experimental Albino Rats.

H1 : Panchanana Rasa will be having Anti-Pyretic activity on experimental Albino Rats.

7. MATERIALS AND METHODS:

7.1 SOURCE OF DATA:

Literary Source :

All the relevent information from Classical texts ,modern text,internet etc. will be reviwed and data will be collected.

Pharmaceutical Source:

Raw material will be collected from the genuine source.

The formulation selected for the present study will be prepared in the P.G Laboratory of

Rasa Shastra , Alva’s Ayurveda Medical College, Moodbidri.

Analytical Source:

Physical and Chemical analysis of prepared formulations will be done in an accredited laboratory.

Experimental Source:

The experiments will be conducted on Albino Rats of either sex that are maintained in standard laboratory, food, water and atmosphere in animal house.The procedure will be condected in Alva’s Ayurveda Medical College,Moodbidri.

7.2.METHODS OF DATA COLLECTION:

A]Pharmaceutical Study :

v  Shodhana of Hingula4 will be carried out.

v  Shodhana of Gandhaka will be carried out5.

v  Samanya Shodhana of Tamra will be carried out6.

v  Vishesha Shodhana of Tamra will be carried out7.

v  Tamra Bhasma nirmana8 will be carried out.

v  Shodhana of Vatsanabha9 will be carried out.

v  Purified and powdered ingredients are added and blended together by triturating with Arka Jala to make formulation10

B] Analytical Study:

The Panchanana Rasa will be subjected for following physico-chemical analysis-

Ø  Organoleptic character

Ø  Loss on drying

Ø  pH value

Ø  Acid insoluble ash

Ø  Acid soluble ash

Ø  XRF

Ø  Relevant instrumental Analysis

C ]Experimental Study:

Samples:

18 Albino Rats of either sex will be selected and divided into three groups

Inclusive Criteria:

a) Healthy Rats of either sex will be considered.

b) Weighing about 150-200g.

Exclusive Criteria:

a) Infected, Pregnant and Diseased Rats

b) Rats which are under trial for other experiments.

Grouping:

Each group contains 6 Albino Rats and will be kept in separate cages.

v  Method: For Anti-Pyretic Study Brewer’s Yeast Inducing Pyrexia Method.

v  Route of administration:

The test drug and made solution with water will be given to control group were administered by oral route with the help of syringes, Yeast suspension will be injected through subcutaneous route.

v  Experimental design :

Pyrexia will be induced by subcutaneous injection of 2g/10ml [20%] yeast suspension in normal saline solution .The drug will be administered after 18th hour of yeast administration .The rectal temperature were recorded by using thermometer [Orchid India] for consecutive 4 hour and 24 hr of yeast administration.

v  Study protocol[method] :

Total 18 Wistre Rats of either sex weighing between 150 g -200 g will be taken and divided randomly into 3 groups, each containing 6 animals. All healthy Albino Rats which were selected for the experiment will be kept under fasting for 18 hrs before the commencement of the experiment .Initial normal rectal temperature of all the animals will be recorded by using a digital thermometer. Fever will be induced by using 20% of Brewer’s yeast solution which will be injected subcutaneously in all Albino Rats in the dose of 2g/10ml body weight and will be placed in the cages kept for them .Then the rectal temperature of each rat will be noted 18th hr after the injection of Brewer’s yeast .After 18th hour of injection of yeast, corresponding test drug will be administered to respective group. Rats of group 1st which will be administered with distilled water served as Control .Animals of group 2nd will be administered with Paracetamol suspension at the dose of 100mg/kg.wt of body weight by using the feeding syringe to serve as reference Standard .Similarly Rats of group 3rd in will be administered with the Panchanana Rasa [Trial group] in a dose of 11.25mg/kg body wt.

After administering corresponding drug to each group hourly rectal temperature of each Rat will be noted for 4 hours and after 24 hours.

v  Statistical test:

As per the data observed statistics will be worked out through one way Annova

test.

7.3. Does the study require any investigations or interventions to be conducted on patients or other humans or animals? If so please describe briefly.

Yes, all the experiments should be conducted on Albino Rats of either sex that are maintained on standard laboratory food, water and atmosphere in animal house.

7.4. Has ethical clearance has been obtained from your institution in case of 7.3?

Yes, clearance will be obtained through the ethical committee formulated in the institution.
