AND ITS ANTIMICROBIAL ACTIVITY”
6. BRIEF RESUME OF THE INTENDED WORK :-
6.1 NEED OF STUDY :
Haratala (Orpiment;Arsenic Trisulphide-AS2 S3) is a compound of
arsenic and sulphur which is yellow Coloured mineral, has been described under Uparas varga in
Rasa shastra it is found in ore form in nature & can also be made artificially. Ayurved text have
three varities of Haratal7 .
i) Patra ,ii)Pinda iii)Tabak.
Patra Haratala is the best amongst all & is used in medicine , It should be in sheets of
golden yellow Colour with luster.Artificially it is prepared by heating arsenic& sulphur together in
anerobic condition. Hartala Bhasma is choisest remedy for various skin disorders .Hartala bhasma
is pungent hot in potency, astrigent in taste, unctuous, & allevates vata& kapha. The study is
intended to purify the good quality of the Patra Hartala, incinerate it ,& then explore the anti
microbial effect of Patra Hartala w.r.t bhasma its Bactriotatic& Bacteriocidal as well as Anti
fungal action against the strains of G ram positive Gram negative Bacteriae & Fungus responsible
for general skin infections.& also to analyse it Physico-chemical properties.This preparation is
made from easily available rawdrugs, economical,& potential in treating the skin disorders.
Considering the above view, my work will be carried out in 3 steps.
1. Preparation of Patra Hartala bhasma.
2. Physico-Chemical analysis of Raw,Shodhita, Marita Patra Hartala.
3. Invitro evaluation if its Bactriostatic ,Bactriocidal &Anti fungal activity
against strains of Gram positive, Gram negative Bacteriae & Fungus
responsible for skin disorders.
6.2 REVIEW OF LITERATURE :-
1. The Grahya Patra Haratala1 should possess the golden colour,it should have glow, bright, shiny,
& should be in flakes form. Such shudha patra Hartala is katu rasatmaka,ushna veerya yukta ,
krimi nashak, kushtaghna, marita patra Hartala is kushtaghna vrana nashaka, rasayana.
Samanya shodhana of Patra Haratala :
1. Grahya Patra Hartala3 is collected ,made into small pieces. It is tied in a pottali.
Kushmand swarasa(Beinancasa hispida) is extracted, Poured in dola yantra & swedana
is done for 3 hrs.
Marana of Patra Hartala :
1. Shodhita Patra Hartala4 is taken & 3 bhavanas of Palasha moola (Butea monosperma)
kwath & one Bhavana of mahish mutra is given Chakrikas are prepared & subjected to
puta using 10 upalas like this 12 putas are given.Thus prepared Patra Hartala bhasma
is used in kushta ,vrana, arsha.
6.3OBJECTIVES OF THE STUDY
1. Preparation of Patra hartala bhasma as per classics.
2. Physico chemical analysis of raw & finished product
3. Invitro evaluation of Patra hartala bhasma Bacteriostatic,Bacteriocidal & Anti FungalActivity
7. MATERIALS AND METHODS :-
7.1 Source of Data :-
1. Sample of Patra Hartala will be collected from the market & get authentificated by the experts
in the subjects.
2. Physico-chemical analysis at a recognised laboratory.
2. The Anti microbial study will be carried out at a recognized laboratory
7.2 METHODOLOGY:
1. The sample of Patra Haratala which is having grahya lakshanas like golden colour , glow,
bright, shiny, flaky, will be collected.
2. Shodhana of Patra Hartala will be carried out according to text.
3. Required quantity of Patra Hartala will be taken ,made into small pieces and tied in a pottali
.Kushmand swaras will be extracted in dola yantra & swedana will be done for 3 hr
. 4. Marana of shodhita patra hartala will be done by using Palash moola kwath & mahish mutra
(Buffalo Urine) as bhavana dravya. Twelve Putas will be given using ten upalas(cowdungs)
every time.
5. Physical changes of the shodhita patra hartala & marira patra hartala will be observed w.r.t
organoleptic characters texture, colour, odour.
Chemical Analysis:
Quantitative method
1. Estimation of percentage of Sulphur .
2. Estimation of percentage of Arsenic.
3. Loss on drying.
4. Total ash content.
5. Acid insoluble ash.
Qualitative method
Trace element analysis.
Instrumental method
Partical size and surface morphology--- by X.R.D method
Antimicrobial Study7: -
Methodology - Microbroth dilution method
Total of 5 micro organisms which included,
Two gram positive bacteria ------Staphylococcus Aureus and E.fecalis
Two gram negative bacteria------E.coli and Pseudamonas
One fungus ------Candida Albicans
will be included to asses the antimicrobial efficacy of the compound The antimicrobial activity will
be carried out by Microbroth Dilution Method. Here serial dilution of the compound will be tested
against fixed density of each bacteria and fungus The results will be expressed asMinimum Inhibitory
Concentration (MIC). After over night incubation , specimen from each tube which shows turbidity
will be inoculated on Blood agar plate. The absence of growth will indicate Bacteriocidal and Anti
Fungal activity of particular concentration of the compound against tested Bacteria and Fungus.
7.3 DOES THE STUDY REQUIRE ANY INVESTIGATIONS OR INTERVENTIONS TO BE CONDUCTED ON PATIENTS OR OTHER HUMANS OR ANIMALS
