INTENDED USE

One of the aims of diary cow feeding management is to improve milk output and shortening the period of no pregnancy.

SPECIMEN: Milk, saliva, blood, urine.

PRINCIPLE

Dairy cow breeder’s aim is to improve milk production and to shorten the non-production period. Breeders desires to have a simple method and in-situ products that they can use to meet this goal that is why, Researchers have develop a simple, easy, effective and cost effective method for rapid diagnosis of cow’s pregnancy. This method can shorten the empty period production and can help breeders to increase breeding, improve economic benefit and to manage the entire breeding cost.

It is based on the changes of progesterone concentration during the cow estrous cycle, the detection of progesterone concentration is to determine whether the dairy cow is pregnant or not.

A glycoprotein hormone called Luteinizing Hormone (LH) is one of the main factors to induce ovulation. The LH in blood, urine and milk will gradually rise before the ovulation. After the LH 12-24 hour peak, ovulation will occur. Pregnancy test kit can reduce the empty pregnancy rate and feeding cost and improve economic growth without any harmful effect. Tracking LH peak can predict ovulation; provide accurate reference data for farmer.

Tracking cow’s ovulation, at the best time to conduct insemination, which can effectively improve cow’s pregnancy rate.

This test can detect ovulation at early stage and can give proper care to cows, sheep and goat according to the corresponding estrus cycle.

SCOPE OF USE

The test is use to detect the early pregnancy stage of dairy cow, goat, and sheep, guiding artificial fertilization at the right time and re-examine of dairy cows/goat/sheep health.

SENSITIVITY

Sensitivity: 100%

HOW TO USE – Cows:
  • Perform the test between 18th AND 22nd post insemination to detect whether the animal is open or is in luteal phase. If found positive then perform one more test on either 23rd OR 24th day post insemination. If this 2nd test also shows positive then the animal’s pregnancy is confirmed. If the 2nd test shows negative then the animal is considered NOT pregnant, even though the test performed between 18th AND 22nd day post insemination is positive. If the first test performed between 18th AND 22nd day shows negative then there is no need to perform this test on either 23rd OR 24th day as the animal is NOT pregnant.
  • If the testing between 18th AND 22nd day post insemination cannot be performed or missed somehow then the test should be performed on BOTH 23rd and 24th day. If the test performed on 23rd day shows positive but shows negative on 24th day then the animal is NOT pregnant. If the test on 23rd day shows negative then the animal is NOT pregnant and there is no need to do the test on the 24th day. If the test shows positive on both 23rd AND 24th day then the animal pregnancy is confirmed.
  • Beyond 24th day and up to 150th day we recommend 5 test in 5 consecutive days to detect pregnancy for the animal whose insemination date is not known of if testing could not be performed between 18th and 22nd OR 23rd and 24th day post insemination. If any one of the 5 tests shows negative then the animal is NOT pregnant and there is no need to perform further test. But if all test performed in 5 consecutive days show positive then the pregnancy is established.

HOW TO USE – Sheep:

The average estrus cycle length is 17 days for sheep. (Progesterone highest peak from 13th day to 17th day after mating with most Progesterone falling between 14 and 20 days for non-pregnant sheep). Perform the test on the 13th day after mating if the test result indicates Positive, do the test again on 15th day. If the test result on 15th day indicates Positive, do the final test on 17th day after mating. If the final test indicates Positive result the sheep is pregnant. If any of the three Test DAYS indicates NEGATIVE result that only means the sheep is not pregnant.

HOW TO USE – Goat:

The average estrus cycle length is 21 days for goats. (Progesterone highest peak from 10th day to 21st day after mating with most Progesterone falling between 18 and 20 days for non-pregnant goat). Perform the ovulation test on the 10th day after mating. If the test result indicates Positive, do the test again on 12th day after mating, if the result on 12th Day test is Positive, do the test again on 14th day after mating, if the test result on 14th day indicates Positive result do the final test on 19th day after mating, if the final test indicates Positive result the goat is pregnant. If any of the four Test DAYS indicates NEGATIVE result that only means the goat is not pregnant.

SPECIMEN COLLECTION AND PREPARATION

Milk: In order to get reliable result, give up the first three milking and then collect sample milk around 5ml.

  • If the Milk is clear, shake the Milk sample to distribute the fats, and directly use for the test.
  • If the Milk is clotted take 1 ml in the test tube, place in centrifuge (10,000 rpm for 10 minutes), in vitro milk was divided into 3 layers, use the micropipette and isolate the 2nd layer of the in-vitro milk sample

Saliva: Collect 5ml Saliva from the mouth and centrifuge at 1500-2000 rpm for 5 minutes. Take the supernatant (2nd layer of the in-vitro saliva sample).

Blood: Collect 1-2ml non-coagulated blood and centrifuge at 2000 rpm for 5 minutes. Let the plasma standby for 30 minutes to separate serum blood clots.

Urine: Use directly.

ASSAY PROCEDURE

1. Remove the test device and milk dropper from its foil wrapper by tearing along the slice.

2. Using the provided dropper, withdraw the sample from the specimen cup and slowly dispense 3 – 4 drops into the circular sample well of the test cassette. The sample liquid moves slowly to the other end due to the capillarity. You will see one or two red lines in the result window.

3. Read results in 5 minutes.

INTERPRETATION OF RESULTS:

Positive: If both T band and C band are visible, the test result is Positive, which means the cow is pregnant or in middle period of estrous cycle.

Negative: If only C line is visible, the test result is negative, which indicates the cow is not pregnant.

Invalid: Only the Test (T) line appears on the membrane, the test is invalid due to improper test procedure or deterioration of reagents. It is recommended that the test be repeated.

PRECAUTION

(1) Test according to the law of the estrous cycle strictly.

(2) For single use only; do not reuse the test devices.

(3) Don’t keep the strip or cassette in the air for a long time after unpacking.

(4) While performing the test, make sure that the surface of sample should not exceed the “MAX” line of strip.

(5) Do not use test kit after expiry date.

VALIDITY AND STORAGE

The period of the validity is 24 months. Store as packaged in the sealed pouch under room temperature. Avoid direct sunlight.
