Specifications forcorrelative Microscopy laser scanning fluorescence workstation
The workstation should comprise latest state of the art technology system with laser scanning confocal microscopy based imaging to meet various challenging and demanding cell biology applications in live cell samples for correlative analysis. The system should be of modular design and upgradeable to higher resolution capabilities in future. Capacity for non-destructive intensity imaging of cell/tissue specimens with special and advanced features for live cell imaging and photo-manipulation procedures.
The system should be offered with the following configuration/capabilities:
1. Fully Motorized Inverted Fluorescence Research Microscope
a.) There should be a motorization of the; Z-drive (including coarse and fine focusing), filter cube movement, revolving objective nosepiece and condenser.
b.) Microscope should be capable of brightfield, DIC and fluorescence imaging.
c.) Motorized Z-focus drive with minimum z-step resolution of 25 nm or better with an active light radiation feedback based controllable system for continuously maintaining focus & ensuring focus stability during fast/long-term experiments (over 72 hrs) along with time lapse data collection for live cell imaging. Motorized autofocus maintenance should be independent of laser wavelengths used for imaging and should encompass IR filter.
d.) Motorized X-Y-Z scanning stage travel range > 100mm x 75mm, resolution better than 1 micron (in X-Y) with joystick control, having sample holder(s) suitable for slides, 35mm glass bottomed petri dishes, Labteck chambers & 24 well tissue culture plates Capability of marking selected observation point and recall along with mosaic/montage scanning.
e.) At least 12V /100w halogen illumination for transmitted light and at least 100-120W metal halide lamp for Fluorescence illumination. The intensity of both illuminations should be controllable. At least 1 spare metal Halide lamps to be included as spares, prices of additional lamps quoted separately.
f.) Motorized DIC attachment with compatible modules and sliders, motorized epi-fluorescence attachment. Six position motorized FL filter wheel, with filter blocks and bandpass filters for the imaging of samples with DAPI, CFP,FITC/GFP,YFP, RFP,TRITC/Texas Red.
g.)Filters for daylight colour balance, heat, neutral density.
h.) At least six position motorized DIC nose piece with following high resolution confocal microscopy grade Apochromatic Objectives / equivalents all suitable for confocal fluorescence imaging, with DIC capability.
(1) ~40X oil DIC at least 1.3 NA.
(2) ~60/63X oil DIC at least1.4NA
(3) ~60/63X oil DIC, TIRF capable >1.4NA
(4) ~100X oil DIC at least1.4NA
i.) Microscope stage environmental control system and growth chamber. Should be provided for the controlling growth conditions of live cells.
Programmable stage mounted incubation system for live cell experiments with easily attachable/detachable incubator. Active CO2and O2controller to maintain 5% CO2hypoxic conditions in the growth chamber. Heating collar & controller for objectives to be part of the system.
Temperature, oxygen and humidity control capability for time lapse imaging for more than 72 hours experiment.
Temperature controller (controllable temperature range between 4 – 40 C or wider range) and humidity controller.
It should be possible to use the motorized stage along with the incubator/environmental control system to acquire LSM images from defined points in the mounted sample(s) which have been selected from online confocal images. Incubator system stage should have suitable insert/holder for slides, LABTECK chambers and glass bottom 35mm Petri dishes.
j.) A suitable anti-vibration table with compressed air floating with an air compressor/pump should be provided for the entire system.
k.) Optional colour digital camera with software control should be provided with a basic resolution of at least 1.4 million pixels.
2. Laser Scan head with FL detectors detection system.
a.) High speed Laser Scanner with a resolution of at least 4K x 4K pixels preferably higher for intensity or spectral imaging. Maximum scan speed of at least 5 frames/sec at frame size of 512x 512 pixels and 1 X zoom, preferably higher and at least 70 frames/sec at frame size of 512 x 32 pixels at 1 X zoom achievable by either single or combination of scanners.
b.) All Fluorescence signals to be confocally collected through computer controlled variable pinhole.
c.) Scan head should be capable of coupling to at least 8 Laser’s with all Laser’s under AOTF control. Fast switching between LASER lines. Scan optics to cover entire wavelength range from ~350nm to 750nm.
d.) Scan field to be at least 18mm diameter or higher.
e.) System should have either at least 4 confocal PMT/hybrid detectors comprising of at least 2PMT + 2 Hybrid detectors. Alternatively two high sensitive spectral detectors (range freely selectable) for sequential detection of upto 4 fluorescence signals and simultaneous DIC brightfield image capture (see point No. h below). In case spectral detectors are offered two additional incident light channels with GASP/hybrid to be provided for simultaneous imaging. Each detector to have independent control and at least 12 bit digitization.
f.) In case of spectral detectors, all spectral detectors should be filter free and able to freely select the fluorescence emission band with a spectral resolution of at least 3nm or better.
g.) Should be possible to combine Confocal PMT detector imaging with hybrid/ detector imaging.
h.) Additional Transmitted light detector for generating transmitted light images in brightfield & DIC mode simultaneously along with fluorescence images in confocal channels.
I.) Scanning capability and data acquisition for X-Y, X-Y-T, X-Y-Z and X-Y-Z-T at multiple points in single experiment along with capability to carry out operations like photoactivation and bleaching.
j.) Laser scanner should havecapability for bleaching/photoactivation & normal scan of any irregular shape to conduct experiments like FRAP, Photoactivation, photo-bleaching, photo-conversion.
k.)Total scan flexibility of line & free hand ROI of multiple shapes and ability to maintain laser intensity for imaging preferred.
l). All scanning capabilities should be accomplished either with single or combination of scanners, Optical zoom at least 1-40X.
m.) Optional capability for fluorescence anisotropy imaging.
3. LASER SYSTEM and Controller Module
Mounted continuous wave LASER sources including fiber coupling to scan head as below:
a.) Blue Diode laser, 30mW or more (preferably higher).
b.) Ar multiline laser, at least 35mW or better, with 458, 488 & 514nm lines, more preferred.
c.) SolidState laser, 561nm at least 20mW.
d.) HeNe/solid state laser, 635/633/640nm ~ 30mW or more power alternatively a combination of 594nm 2mW and 633/635/640nm 5mW laser.
f.) Lasers to be optically coupled and connected to scan head via fiber optics through multi channel AOTF.
g.) AOTF for control of all offered lasers for fast attenuation and switching in synchronization with scanner of independent laser lines simultaneously.
4. System control computer workstation
Latest PC configuration with high end Xeon processor or better to be supplied at time of shipment. Tentatively greater than 2.9 GHz clock speed, 16GB (preferably higher) DDR2 RAM, DVD writer, 300GB HDD. High resolution graphics card. Gigabit Ethernet, USB, Firewire port, 30” or at least 24” x 2 LCD TFT monitors. Professional Windows 7, 64 bit English version.
5. Software
a.) Software should be provided to control all parameters of image acquisition & measurements/analysis including laser scanning, detection, and microscope control. The system should be able to process and analyze images and carry out operations like, multiple position marking and recall, intensity measurements across a line, basic image analysis measurements of area, perimeter, pixel intensity, etc.
b.) Software should be able to carry out 3D reconstruction from Z stack series, 3D animations, rendering of 3-D surfaces 3D reconstruction & animation.Image Deconvolution (including 3D Deconvolution), Module for complex 3D time series experiments (acquisition, compilation, analysis & presentation). Topography analysis & visualization of 3-D surfaces along with 3-D measurements. Capability for Montage /Mosaic images, extended depth of focus display from Z-stack images acquired in X-Y-T, X-Y-Z-T series, image stability and handling for long term experiments. Data organization capabilities.
c.) Should be able to carry out ROI bleach for FRAP, FLIP,Photo-activation/conversion.Extensive monitoring,recording and analysis software for long term time series images withgraphical means of ROI analysis.
d.) Capability of recording and evaluation of acceptor photobleaching as well as sensitized emission FRET experiments.Recording of FRET image data with subsequent evaluation supporting both the methods of acceptor photobleaching as well as sensitized emission. FRAP experimentswith subsequent analysis of the intensity kinetics.
e.) Software should be capable of time lapse imaging with maintenance of focus conditions. The software should be able to carry out physiology applications like real time emission intensity measurements and graphical mean of ROI analysis for applications like calcium measurements and monitoring changes of marker levels in live cells.
f.) Software should also be able to carry out statistical operations like colocalisation analysis and pixel frequency distribution.
6. Installation and service supports
a.) Complete installation and on site training by companies engineers & application experts to be provided.
b.) Bidder should include the supply of appropriate Online UPS system to support the complete system including lasers from a standard branded manufacture. Wiring and connection of UPS to system to be carried out as part of installation.
c.) Appropriate ergonomic furniture/accessories (system table, stands, switches, cords, connectors, extension boards, etc.) to be provided to make complete workstation fully functional for operation.
d.) Two years comphrensive Warranty on complete system (including lasers) with additional one year Service warranty to be offered on complete system.
e.) Bidder should also provide a detailed list of users of the quoted system in India with contact details.
f.) Bidder should provide all pre-installation requirements to have the system installed in suitable room conditions.

TENDER DOCUMENT FOR OPEN/GLOBAL TENDER
(TERMS & CONDITIONS INCLUDING INSTRUCTION TO BIDDERS AND CONDITIONS OF CONTRACT)
- GENERAL: Quotations in closed cover are invited on behalf of the Director, INSTITUTE OF MICROBIAL TECHNOLOGY, Chandigarhfrom the reputed Indian and foreign manufacturer. The offer/ quotation must be strictly as per required specifications and the tender terms & conditions.
- PREPARATION AND SUBMISSION OF OFFERS:
a.Quotation should be submitted directly by the original manufacturer/supplier or its sole authorized distributor/dealer. In case of bid by authorized dealer/distributor, the manufacturer authorization should be attached with the technical bid as per Annexure-‘G’.
b.In case a bidder is not doing business within India, it shall furnish the certificate to the effect that the bidder is or will be represented by an agent in India equipped and able to carry out the supply, maintenance, repair obligations etc. during the warranty and post-warranty period or ensure a mechanism at place for carrying out the supply, maintenance, repair obligations etc. during the warranty and post-warranty period.
c.The bidder shall bear all costs associated with the preparation and submission of its bid irrespective of the conduct or outcome of the bidding process.
d.The bidder should not indulge in any corrupt, fraudulent, collusive, coercive practices during the entire process of procurement and execution of contract/order.
e.Before the deadline for submission of the bid, IMTECH reserves the right to modify the bidding document and to extend or not to extend the date of submission. Such amendment/modification will be notified to bidders receiving the bidding document in writing or by cable/fax/ email and will also be hosted on IMTECH website.
f.Conditional tenders will be summarily rejected.
g.A bidder may withdraw, substitute, or modify its bid after it has been submitted by sending a written notice signed by the authorized signatory before the date of submission of the bid. Any re-submission or modification in the bid should be submitted before the date & time of submission of bid as originally conveyed in the invitation of bid.
h.No bid may be withdrawn, substituted or modified in the interval between the deadline for submission of bids and the expiration of the period of bid validity specified by the Bidder on the Bid Form or any extension thereof.
i.First envelope marked “WITHDRAWAL” shall not be opened, but returned to the Bidder subject to submission of valid authorization to request the withdrawal. In case of substituted and modified bid, only the substituted bids and modifiedbids will be opened subject to production of authorization from the bidders.
j.The quotation should be addressed to the Director, INSTITUTE OF MICROBIAL TECHNOLOGY, Chandigarh and sent to the Stores & Purchase Officer.
k.Each offer/quotation should be kept in separate envelope against each item of the tender notice.
l.For items covered under Two-Bid -Tender System, quotation/offer should be submitted in two separate envelopes containing Techno-Commercial bid and Price bid and these put in one envelope.
m.The outer cover containing the offer/quote should be super-scribed with our Tender No. (Not global/open Tender Notice No), Due date and Date of opening of tender.
n.The offers must containthe following documents :-
(A)Techno-commercial offers must contain:
- Manufacturer authorization (as per clause 2(a)) as per Annexure –‘G’.
- Certificate by bidder not doing business in India as per clause 2(b).
- Technical literature/ leaflets and complete specifications of quoted model(s) along with commercial terms and conditions.
- Compliance statement/questionnaire of tender terms and conditions as per Annexure-‘A’
- Compliance statement of specifications as per Annexure- ‘B’
- Bid Security/EMD (as per clause -4) as per Annexure- ‘C’
- In case of exemption from submission of Bid security, proof of registration with DGS&D/NSIC as per clause (4(a)).
- Integrity Pact- Only in case of bids of more than Rs. 3.00 Crore (as per Annexure- ‘D’)
- Copies of previous supply orders (as per clause 10(c) and clause 25) as per Annexure –‘H’.
- Details of supplies of similar equipments as per clause 10(d).
(B)Price Bid offers must contain: (Only in case of two bid system)
i.Bid price as per clause 5 in format enclosed as Annexure- ‘E’ or Annexure- ‘F’ as applicable.
ii.Certificate of price reasonability as per clause 10(b).
- DUE & OPENING DATES: The Offer/Quotations must reach at IMTECH as mentioned above on page No.1in the presence of bidders, who wish to present themselves at the time of opening of tender. In case opening date happens to be a holiday, the tender will be opened on next working day at the same time & location. The price bids of two bid tender system shall be opened after technical evaluation of technical bids. The date of opening of price bids shall be informed to the bidders found suitable in technical evaluation. No request for extension in the date of submission & opening of tender will be entertained. However IMTECH, may at its discretion, extend the deadline for submission of bids under intimation to bidders.
- EARNEST MONEY DEPOSIT (EMD):
- EMD in the form of Bank guarantee (As per format enclosed as ANNEXURE- ‘C’) or Term Deposit Receipt/FDR or Bank Draft of a scheduled bank pledged in the name of Director, INSTITUTE OF MICROBIAL TECHNOLOGY, Chandigarh valid for 180 days from the date of opening of the tender for the required amount as mentioned in the tender notice separate for each item must be submitted along with the quotation (with the techno-commercial quotation in case of two-bid system and photocopy of the same enclosed with the price bid and for foreign manufacturers they should furnish the EMD in foreign currency equivalent to Indian rupees,otherwise quotation may not be considered. The firm registered with DGS&D/NSIC as manufacturer for the supply of the same category of item for which the party is submitting quotation will be exempted from submission of EMD. Intended parties will have to give proof of registration along with their quotation. EMD of the unsuccessful bidders shall be refunded without any interest at the earliest after finalization of the purchase of concerned item. The party must therefore, submit a pre-receipted Bill in triplicate along with the quotation (in case of EMD sent in form of Bank Draft) to enable us to refund their EMD.
- The bid security /EMD may be forfeited if a bidder withdraws or amends or impairs or derogates its bid during the period of bid validity.
- PRICES
Bid prices should be filled in the appropriate format enclosed as Annexure ‘E’ and ‘F’.
- For goods manufactured with India:
(i)The price of goods quoted Ex-Works including taxes already paid.
(ii)VAT and other taxes like excise duty etc. which will be payable on the goods if the contract is awarded.
