Description
BluePlus™ Protein Staining Solution is a ready-to-use solution for a highly sensitive staining of proteins separated in polyacrylamide gels. BluePlus™ contains Coomassie Brilliant Blue G-250 dye. Due to the property of the dye to form colloidal particles, proteins are preferentially stained without significant staining of the gel matrix. The linear dynamic range of BluePlus™ extends over two orders of magnitude (see picture below) which is superior to Coomassie R-250. The stain does not contain toxic methanol or acetic acid.

Features
·  Ready-to-use.
·  Safe - does not contain methanol or acetic acid
·  Rapid staining procedure, no destaining required
·  End point stain, overstaining does not occur
·  More sensitive than Coomassie Brilliant Blue R-250
·  Linear dynamic range from 5 ng to 500 ng
·  Compatible with mass spectrometry and silver staining
·  Economical - can be reused up to three times

Sensitivity
Detects as little as 5 ng of protein.

Storage
Store at 4 °C

Sensitivity and linear dynamic range of PageBlue™ Protein Staining Solution.
a - beta galactosidase,
b - bovine serum albumin.
Electrophoresis conditions: 12% SDS-PAGE, 0.75mm gel, well width 3mm.

Staining Protocol for Mini-gels
Fast Protocol:
1.  Washing (only for SDS containing gels):
·  Place the gel into a tank and add 100ml of deionized or distilled water.
·  Put the uncovered tank with the gel into a microwave oven and turn on high power for 1 min. Do not boil.
·  Take out the tank from the microwave oven and continue washing with gentle agitation for 7 min.
·  Discard the wash.
Repeat 3 times. / 3x8 min
2.  Staining:
·  Add 20 ml (or enough to cover the gel completely) of BluePlus™ Protein Staining Solution.
·  Put the uncovered tank into a microwave oven and turn on high power for up to 30 seconds. Do not boil.
·  Take out the tank from the microwave oven and continue staining with gentle agitation for 20 min.
·  Discard the staining solution. / 20 min
3.  Washing (optional):
·  Add 100ml of deionized or distilled water and rinse for 5 min.
·  Discard the wash and add 100ml of deionized or distilled water for gel storage. / 5 min
Total time: / 25min for native gels;
40 min for SDS containing gels
Conventional Protocol:
1.  Washing (only for SDS containing gels):
·  Place the gel into a tank and add 100 ml of deionized or distilled water and rinse for 10 min.
·  Discard the wash.
Repeat 3 times. / 3x10 min
2.  Staining:
·  Add 20 ml (or enough to cover the gel completely ) of BluePlus™ Protein Staining Solution and staining with gentle agitation.
·  Discard the staining solution. / 60 min
(or overnight)
3.  Washing:
·  Add 100ml of deionized or distilled water and rinse for 5 min.
·  Discard the wash and add 100ml of deionized or distilled water for gel storage. / 5 min
Total time: / 65min for native gels;
95min for SDS containing gels
Note:
·  The staining solution can be reused up to 3 times without noticeable decrease in sensitivity
·  Large gels require more staining solution, as gels should be completely immersed in it. When several gels are stained, increase the amount of staining solution accordingly.
·  Washing (step 1) is dedicated to the removal of SDS from the denaturing gels since SDS interferes with the staining. This step can be omitted for native gels.
