The EXPORTER has developed a novel version of batch chromatography utilizing proprietary nanotechnology (the TECHNOLOGY). The power of this technology is that, with no specialized equipment, biomolecules of research, clinical or forensic interest can be captured from a large, dilute sample and then enriched and concentrated. The EXPORTER is developing a suite of simple-to-use products for the manipulation, capture, purification, and concentration of DNA, RNA and proteins.
The TECHNOLOGY is based on high-density nanoparticles used as a platform to support a family of high-value genomics and proteomics applications. These nanometer size particles form extremely stable suspensions with enormous surface area in biological fluids. The EXPORTER has exploited the unique physical properties of nanoparticles in suspension, and has coated these particles with biomolecule-specific films to create a new class of tools for the biotechnology marketplace. The resulting nanoparticles have the unique ability to capture and concentrate specific biomolecules from large or extremely dilute solutions. The biomolecules bound to these nanoparticles can then be released in a purified and concentrated form ready for further biochemical or genetic testing. This extraction-capture-concentration process easily scales from liter size samples to samples as small as a 1/100th of a milliliter by a simple adjustment of the proportion of the nanoparticles used in the preparation. The technology can process samples with as little as 100 picograms of DNA.
The EXPORTER is working with a number of high-profile academic and governmental laboratories to establish the TECHNOLOGY as the new standard in sample processing for a range of genetic and forensics applications. A complementary suite of the EXPORTER'S RNA and protein products, to support biomarker discovery and molecular epidemiology, are in development. As a new generation of DNA, RNA, and protein biomarkers become ready for personalized medicine and gene-based disease risk assessment, the EXPORTER'S TECHNOLOGY will provide simple, rapid and field-ready tools for the capture, enrichment and stabilization of those biomarkersto a microliter-sized preparation ready for detailed molecular analysis.
The KIT and future EXPORTER products are distinguished by:
•their extreme simplicity of use,
•their cost effectiveness,
•the enormous binding capacity of the nanoparticle matrix,
• rapid binding kinetics associated with nanotechnology processes, and
• the batch protocol is a "one-pot process"; that is, the whole extraction process, from start to finish, is performed within one low cost standard microfuge tube.
•The EXPORTERS products can process the smallest and rarest biological samples. These include micro-dissected clinical biopsies, crime scene wipes, clinical skin swabs, and clinical samples such as bacterial swabs.
•The TECHNOLOGY is readily scalable in sample size from a few microliters to liters.
A few cells in 10 microliters at one extreme, or microbial DNA in a liter of waste water filtrate at the other.
•The EXPORTER'S products are scalable in sample content. The PrepParticle MicroKit can process DNA samples that contain 100 picograms up to 15 micrograms. A 150,000 fold range!
•The TECHNOLOGY can be modified, with fine-tuning, for DNA, RNA, Protein and Microbes. Several spin-off products are in development.

Product Profile--the "KIT"

The KIT utilizes ceramic nanoparticles and a simple to use, "one pot" process to capture, purify, concentrate, and desalt of DNA from samples in the 50 to 500 microliter range. The KIT requires ordinary microcentrifuge tubes, a standard micropipettor, and an inexpensive bench top centrifuge. All of the DNA processing steps are performed at room temperature and can be done manually. The KIT protocol can be further adapted to standard laboratory automation. Retail pricing starts at US$65 for a 50-sample KIT. KIT applications:

Purify DNA from
• Fresh & frozen blood samples in the 100 uL to 0.05 uL range
• Thin-sections of fixed, paraffin embedded tissue.
• Forensic samples from surface wipes, such as blood, semen, and other bodily fluids.
• Dried blood extracted from paper cards such as FTA or Guthrie.
• Buccal cell samples, from either swabs or mouth rinse.
Concentrate and de-salt the DNA from
• DNA purification processes including column or batch chromatography (spin columns or magnetic beads) and organic extraction and
• Nucleic acid amplification reactions including PCR or whole genome amplification.
