Plasmid pRc/CMV-HBs(S), usually called pCMV-S. 5618 b.p.
The HBsAg coding region ORF is from positions 939 to 1619 bp
681 bp, or 226 AA plus Stop codon).
To prepare pCMV-S, the HBV sequence was removed from a Bluescript vector by KpnI-BssHII digestion. This fragment was then cloned into the corresponding sites of the pRc/CMV expression vector (Invitrogen). The KpnI site is in the pRc/CMV vector's polylinker, just after the CMV promoter, and the BssHII site is found at the end of the vector's NeoR gene. This cloning step therefore removed the pRc/CMV vector's original bGH sequence, f1 origin, SV40 promoter, and most of the NeoR gene. The HBV sequence contains its own polyadenylation signal, which is then followed by the pRc/CMV vector's SV40 termination site. (Robert Whalen, Paris, France, March 1998)
