6.1 Screening of genotypes for high-temperature tolerance at seedling stage.

Fortysix genotypes of Indianmustard (B.juncea)were tested for high-temperature tolerance at seedling stage under field and laboratory conditions. 100 seeds of each genotype were sown in 2m row length on 23rd September 2015 at Hisar and Kanpur while on 2nd October 2015 at Ludhiana with three replications.The experiment was continued for 30 days.The same set of genotypes was evaluated in laboratory and for this the genotypes were sown in plastic trays containing 5 kg soil at optimum moisture conditions required for germination. Sowing was done in rows with three replications for each genotype in random order to avoid minor microclimatic variation inside the tray and seed germinator. Seedlings were initially allowed to grow at 250C± 10C for 5 days at 70-80% RH and then exposed to 450C±10C at 30% RH for 4 hrs (heat shock treatment) daily, for 4 days. Percent seedling mortality and dry weight of 10 seedlings were recorded under field and laboratory conditions. Higher seedling mortality indicated higher susceptibility of the genotypes to high temperature and vice-versa.

Seedling mortality and dry matter recorded at Hisar, Kanpur and Ludhiana varied significantly within the genotypes.

Hisar|

Seedling mortality ranged from 16.5% in Kranti to 100% in PDZ-1 and KMR (L) 1506 in thelaboratory. Maximum dry weight was observed in Kranti (73.5 mg) followed by DRMR1165-40(71.0 mg) while minimum dry weightwas inPDZ-5 (29.4 mg).The maximum temperature recorded was 38.2°C with anaverage of 35.4°C and minimum temperature was 13.5 °C with anaverage of 19.8 °C under field conditions.Seedling mortality ranged from 20% in KMR (E)15-2 to 100% in PDZ-1 and KMR (L)1506.Based on low seedling mortality and high dry weight the genotypes RH 0555, Kranti and DRMR1165-40 wererated asheat tolerant at seedling stage

Kanpur

Seedling mortality varied from 14.5 (LES-49) to 43.0% (DRMR15-9) and seedling dry weight from 29.5 in RK(E)8-2 to 49.0 mg in Pusa mustard 25 among the tested genotypes under laboratory. Low seedling mortality (<20%) and high seedling biomass (>40.0 mg) rated genotypes RH1301, Pusa mustard 25, PRE2012-14,PRE2011-6, PDZ-4, NPJ-197, NPJ-196,NPJ-195, NPJ-194,LES-53,LES-50, LES-49 and KMR15-3 as high temperature tolerant at seedling stage. Average temperature varied from 24.2°C to 28.4°C during the experimental period. Seedling mortality ranged from 14.7 (PRE2011-6) to 44.7% (PDZ-2) and seedling biomass varied from 12.5(RK (E)8-1) to 28.1 g (NPJ-194 and LES-50) in field sown genotypes. Genotypes Pusa mustard25, PRE2011-6, NPJ-197, NPJ-196, NPJ-195, NPJ-194, LES-53, LES-50, LES-49, KMR(E)15-1 and DRMR15-9 were characterized as high temperature tolerant based on seedling mortality <20% and seedling biomass >25g.Considering low seedling mortality and high seedling biomass eight genotypes i.e.Pusa mustard25, PRE2011-6, NPJ-197, NPJ-196, NPJ-195, NPJ-194, LES-53, LES-50 and LES-49 were identified high temperature tolerant at seedling stage under both the conditions.

Ludhiana

Seedling mortality varied from 14.3% in PRE2011-6to45.3% inDRMR15-9while dry matter/10 seedlings ranged from30.2 (RK(E)8-1) to 56.2 mgin DRMR1191-2. Seedling mortality of <20% and dry matter of 10 seedlings >40mg identified ninegenotypes viz.PRE2012-14,PDZ-4,NPJ-197, NPJ-196,NPJ-185,NPJ-194,LES-53,LES-50 and LES-49as thermotolerant at seedling stage. Genotype DRMR15-9 succumbed to heat shock with seedling mortality >45% and was rated as susceptible at seedling stage. The maximum temperature recorded was 34.2°C with anaverage of 27.1°C and minimum temperature was 14.8°C with anaverage of 22.7°C. In thefield,sown genotypes mortality ranged from 5.6% (DRMR1165-40) to 60% (NPJ-198) while the dry matter varied from 9.6 (LES-53) to 56.1g in NPJ-190.Genotype DRMR1165-40 showed lower mortality and high dry matter .With mortality (<20%) and dry weight>25 g only two genotypesDRMR-4104 and DRMR1165-40 were identified as thermotolerant under field conditions while RGN-337,NPJ-197,KMR15-3,DRMR-4104,DRMR-4001,DRMR1617-45,DRMR15-9,DRMR1165-40and 45S35 registered mortality <30% and dry weight25 g categorizing them to be moderately tolerant

Overall mean at the threecenters indicated minimum seedling mortality of 18.2 % (LES-53) and maximum of 49.6 %(RK(E)8-1 and 45S35) while dry weight of ten seedlings ranged from 31.2(PDZ-2)to54.7 mg (PRE2012-1).Under field conditions, the overall seedling mortality was minimum in NPJ-194(25.2%) and maximum in PDZ-1 (53.6%) at three locationsand dry matter varied from 14.6 (PRE2012-14) to 39.3g (DRMR1165-40)at two locations.Genotype Kranti at Hisar and Kanpur, 45S35at Hisar and Ludhiana andDRMR15-9 at Kanpur and Ludhiana were rated as thermotolerant.

6.2 Evaluation of genotypes under low light stress condition

Five genotypes were sown in paired rows atrow spacing of 30cm in the field with 3 replications under RBD. Treatments comprised i) shading with nets that cut 30% natural light and ii) no shading (control). Shading was given for 30 days from mid-December to mid-January. All the recommended practices were followed to raise a healthy crop. Physiological traits like leaf water retention (LWR), specific leaf weight (SLW), relative water content (RWC) and SPAD were measured in each genotype per replicate 10 days after removal of nets/shading treatment using thestandardized procedure.Yield and yield components were recorded at maturity.

Shading reduced physiological traits likeSPAD,LWR and SLW to anappreciable extent. The impact of shading was significant on yield attributes and yield at Kanpurand Ludhiana.

Kanpur

Physiological traits like SLW varied from6.5 (RH-749) to 7.8 mg/cm2(Kranti) under control andfrom 5.3(Kranti) to 6.2mg/cm2(DRMR541-44) with 30% shading. Reduction in leaf water retention was <10% in all the genotypes. SPAD values varied from 42.7 (BPR349-9) to 46.5 (Kranti) under natural sunlight and from 37.5 (BPR349-9) to 41.8 (DRMR541-44) with shading treatment. RWC declined by less than 10% in all the genotypes (Table 6.2.1). Mean SLW declined by18.5%, LWR by 6.3%, SPAD values by 11.0% and RWC by 5.6% with shading treatment. Reduction in total siliquae per plantwas <20% inDRMR541-44 and RH-749.Shading impaired seed weight by 4% in DRMR541- 44 andRH-749.Again seed yield was reduced by <20% in DRMR541-44 (16.3%) and RH-749 (18.3%).Mean decline in siliquae per plant (32.0%), seed weight (12.2%) and seed yield (27.4%) was registered with shading in comparison to control.Genotypes DRMR541-44and RH-749showed <20%decrease in seed yield and were identified as promising strains under low light stress.

Ludhiana

Differences existed among genotypes for physiological traits under natural light conditions(Table6.2.1).Shading affected physiological traitsexcept SPAD and RWC to avariable extent in the genotypes even 10 days after removal of nets. Mean decline was 41.6% in SLW, 6.8 % in LWR,2.5% in SPAD and 4.6% in RWC over unshaded condition. Genotypes possessed higher physiological traits under control/natural light and showed only less than 10% decline in LWR except in RH-749.Siliquae per plant varied from 232.5 to 295.2 under natural light and from 137.8 to265.0 under shaded conditions with a reductionless than 10% in DRMR541-44 and RH-749.Seed weight ranged from 3.8 to 5.7 g under control and 3.4 to 5.0 g under shaded treatment. Only DRMR541-44 registered <10% decrease in seed weight over unshaded/control(Table 6.2.2).Shading reduced a mean number of siliquae /plant by 23.3%,seed weight by 13.0% and seed yield by 37.4% over control.Only genotype NRCHB-101(16.8%) recorded significantly lower reduction in seed yield while most of the genotypes suffered yield reduction >25%

Based on thelesser reduction in physiological traits and seed yield (<20%) genotypesDRMR541-44 andRH-749 at Kanpur whileNRCHB-101at Ludhianawere identified as promising under low light stress.

6.3 Screening of genotypes for drought tolerance

Fortysix genotypes of B.juncea were evaluated for drought tolerance under field conditions at Hisar, Kanpur, and Ludhiana. Each genotype was sown in 5 rows with

2.5mrow length. The test genotypes were sown at 30 cm row spacing after pre-sowing irrigation.Treatmentswere replicated 3 times under RBD factorial design. Main treatment comprised, i) two post sowing irrigations at 35 and 65 DAS and ii) no post sowing irrigation.Analysis of variance revealed significant genotypic differences for RWC,SPAD values, yield components and seed yield.

Hisar

Chlorophyll content was measured in terms of SAPD values. Out of 46 genotypes, 25 showed <10% reduction in SPAD values under rainfed condition with anaverage reduction of 11.2% (Table-6.3.1). Maximum reduction was 22.6% in LES-49 and aminimum of 2.5% in DRMR15-16. Thirtytwo genotypes showed <10% decline in RWC under rainfed over irrigated conditions. The maximum decline was in DRMR541-44 (34.4%) followed by NPJ-198 (28.3%) while the minimum reduction was in DRMR10-40 and NPJ-179 (1%).Twenty genotypes showed <10% reduction ina number of siliquae on themain shoot (SMS) under arainfed condition with anaverage reduction of 11.4 %. The minimum reduction was recorded in PRD2013-2(2.0%) followed by DRMR1187-71 (2.1%). Twentyseven genotypes showed <10 % reduction in a number of seeds/siliqua under rainfed situation with the average reduction of 9.4%. 1000-seed weight also declined under rainfed condition and 31 genotypes showed <10% reduction out of them 13 genotypes showed <5% decline. The average reduction was 8.6% and with a minimum in KMR1-4 (0.07%) followed by NPJ-197 (1.2%). Biological yield ranged from 63.3 (PDZ-1) to 180.0 g/plant (LES-53) in the irrigated condition and 41.0 (PDZ-5) to 167.5 g/plant (LES-53) under the rainfed condition. The mean reduction in biomass was 32.0% under rainfed over irrigated conditions. Thirteen genotypes recorded less than 20% reduction in biological yield with the minimum reduction of 6.3% (PRD2013-2) followed by 6.9% (LES-53) and maximum reduction in PDZ-5 (67.5%). Seed yield/plant ranged from 10.9 (PDZ-1) to 54.5g/plant (LES-53) in the irrigated and from 7.3(NPJ-182) to 44.3 g/plant (LES-53) under rainfed conditions. Mean seed yield declined by 39.4% under moisture deficit. Eight genotypes showed <20% reduction in seed yield/plant with aminimum of 11.5% in PRD2013-2 followed by 14.1 % in NPJ-197. Twentynine genotypes showed < 10 % reduction in harvest index with amean reduction of 10.9% over irrigated condition. Oil content was reduced to 2.6% under stress. Maximum oil content of 40.9% and 39.3% was in NPJ-182 under irrigated and rainfed conditions respectively. Seven genotypes showed ≤0.5 DSI and 16 genotypes showed DSI value between 0.5-1.0. Genotypes LES-53, PRD2013-2, DRMR4001, PDZ-1, RB50, NPJ-197 and KMR (E)15-1were found tolerant under rainfed condition on the basis of lesser reduction in seed yield, harvest index, seed weight,biological yield and also DSI ≤0.5.

Kanpur

Mean reduction in SPADchlorophyll values was 12.5 % and RWC 31.7% (Table 6.3.1). Less than 10% decline in SPAD chlorophyll content was reported in 22 genotypes. RH0673 registered comparable SPAD values under both irrigated and rainfed conditions with minimum decline of 0.3%.None of the genotype registered <10% decline in RWC under rainfed condition while thedecline in RWC was to the tune of 15.1% in PDZ-6 followed by16.9% in DRMR1153-32.These two genotypes registered aminimum reduction in RWC. Siliquae on themain shoot was reduced to 17.1% over irrigated condition (Table 6.3.2).Less than 10% reduction for this trait was reported in fifteen genotypes. Siliquae on themain shoot were comparable in RH 406under both irrigated and rainfed conditions.Seeds per siliqua were reduced by 9.1% under rain fed condition. 33genotypesshowed <10% decline in seeds/siliqua. Seeds per siliqua were comparable under both irrigated and rainfed conditions inPDZ-1, PDZ-2 and PDZ-5. The meanreduction in 1000seed weight was 15.2% under rainfed condition. Only 9 genotypes maintained higher 1000seed weight with <10% reduction as compared to the irrigated module (Table 6.3.3).Biomass ranged from 55.8 (RGN-337)to 88.5 g/plant (PDZ-6)under irrigated and from 45.3 (DRMR541-44) to75.9g/plant(PRD2013-2)under rainfed with a mean decline of 32.0%.Seed yield varied from 11.1 (DRMR541-44) to 20.3 g/plant (DRMR-4104 and PDZ-6) and from 7.8(DRMR541-44) to 16.8 g/plant(NPJ-198) under irrigated and rainfed conditions respectively (Table 6.3.4).Harvest index declined by 10% in 19 genotypes and < 5% in 7 genotypes with mean reduction of13.2 % under stress .Seed yield was reduced by 24.6% under moisture stress. Yield reduction was <20% in 12 genotypes with DSI≤1.0and in 6 genotypes with DSI≤0.5.

Ludhiana

The mean reduction in SPAD values was 13.3% and RWC was 7.9 % under moisture stress over irrigated module.Twentynine genotypes maintained higher RWC as revealed by less than 10% decline in this trait (Table 6.3.1). Minimum reduction in RWC (<1.0%) was recorded in genotypeLES-50,DRMR-4001,RB50,DRMR10-40 and RGN-368. Fourteen genotypes showed <10% decline in SPAD chlorophyll values. The minimum decline in chlorophyll (0.4%) was in PDZ-5.GenotypesLES-50,RB 50 and RGN-368 had comparable values of RWC while PDZ-5exhibited for SPAD chlorophyll under both irrigated and rainfed regimes. The mean reduction in siliquae on themain shoot was 12.1% over irrigated module. Less than 1% decline in KMR(E)-1505 (0.3%) trailed byDRMR10-40 and DRMR1165-40 (0.8%) was recorded for this trait (SMS). Twentyfive genotypes showed <10% decline in siliquae on themain shoot with the

imposition of moisture stress. Less than 10% decline in seeds/siliqua and seed weight has been recorded in twenty andthirteen genotypes respectively.The mean reduction in seeds/siliqua was 12.2% and seed weight 15.6% under rainfed condition compared to irrigated condition (Table 6.3.3).Minimum reduction in seeds/siliqua has been recorded in LES-50(0.4 %)followed byNPJ-195 (0.9%) and for seed weight in NPJ-194(0.3%). Biomass varied from 3768.9 (DRMR15-47) to 7600kg/ha (NPJ-198) under irrigated condition and 3173.3 (RH 406) to 6693.3kg /ha (NPJ-195) under rainfed module with a mean reduction of 13.1%.Biomass declined by <10% in twentytwo genotypes and by <20% in thirtyfive genotypes.Seed yield varied from 518.2 (LES-50) to1706.7kg /ha (Pusa mustard 25) under irrigated and from 470.2(LES-50) to 1332.4kg/ha(Kranti) underrainfedregime with a mean decline of 13.5% (Table6.3.4).Thirty sevengenotypes showed <20% decline in seed yield and out of them twentythreegenotypes hadDSI<1. Twentyone genotypesregistered10% reduction in yield with DSI≤0.5 in fourteen genotypes only. Mean HI (%) was 20.8 and 17.8 under irrigated and rainfed regimes (Table 6.3.4).Oil content ranged from 36.4 (LES-52) to 40.8% (DRMR-1165-40)) under irrigated and from 36.0 (NPJ-182) to 39.7%(DRMR-15-14) under rainfed conditions.

Based on seed yield reduction of <20% and DSI≤1, only one genotype LES-53 at Hisar and Kanpur,DRMR15-5,PDZ-2,DRMR10-40,NPJ-179, LES-52,RH-1301 at Kanpur and Ludhianaand DRMR-4001,RB50,NPJ-197and KMR(E)15-1 at Hisar and Ludhiana showed tolerance to moisture stress. GenotypePDZ-2 and DRMR10-40 atKanpur and Ludhianaand again only three genotypes viz. DRMR-4001, RB50, and KMR(E)15-1 at 2 locations (Hisar and Ludhiana) showed yield reduction <10% with DSI≤0.5 and were rated as highlytolerant to moisture stress.
