Phase Contrast

Initial setup

1. Follow ALL steps of Köhler Illumination

2. Move <10x +> objective into OP

3. Turn the Condenser Turret to put the proper Phase Annulus in the OP

a. 10x,20x = Ph1

b. 40x=Ph2

4. Turn aperture diaphragm to the right to open fully

a. If this is not fully open, the OP will be restricted reducing the contrast

5. Raise the field diaphragm lever to open the field diaphragm fully

Center the Annular Diaphragm

6. Set the eyepiece tube turret to <B>

7. If the rings are not aligned…

a. Call or email Justin

b. DO NOT ATTEMPT TO ADJUST THIS

8. Move eyepiece tube turret back to <O>

Conduct Observation

9. Make sure aperture diaphragm is still fully open

10. Lower the field diaphragm lever so that the field diaphragm is just wider than the field of view

11. Change filters

a. Remove NCB filter

b. Use ND and D filters to adjust brightness

i. DO NOT ADJUST THE BRIGHTNESS BY TURNING THE BRIGHTNESS ADJUSTMENT DIAL – should be set to <12V100W>

c. Move GIF filter into OP

i. Improves contrast – yes, this will look green
